AI Analysis of 400,000 Reddit Posts Flags Overlooked GLP-1 Drug Symptoms

A University of Pennsylvania team used AI-assisted computational social listening to analyze more than 400,000 Reddit posts written by nearly 70,000 users over more than five years. The study examined reports involving semaglutide drugs, including Ozempic, Wegovy and Rybelsus, and tirzepatide drugs, including Mounjaro and Zepbound. Researchers used large language models to help map informal descriptions to standardized adverse-event terms such as those in MedDRA. About 44% of users in the dataset mentioned at least one side effect, with gastrointestinal complaints the most common and consistent with established drug effects. Nearly 4% of users who reported side effects discussed reproductive symptoms, while fatigue was the second most frequently reported complaint despite being less prominent in many clinical trials. The findings show associations in online discussions and do not prove that the drugs caused the symptoms. Reddit users are not representative of all patients, tending to be younger, more likely to be male and disproportionately based in the United States. The researchers say the patterns, potentially relevant to the hypothalamus’s roles in hormone and temperature regulation, should be tested through controlled research. They plan to examine other platforms and non-English communities to assess whether the signals generalize.
